Registration Date 12-02-2021 Organization Selection-Genetic Institute - National Center of Seed and Selection Studies of Ukrainian Academy of Agrarian Sciences popup.description2 As results F3’5’H-In gene polymorphism and Satt229 and Satt006 gene conservation were determined. in a sample of 159 nucleotide sequences from EMBL and NCBI databases. The polymorphism of 38 soybean varieties at loci F3’5’H-In, Satt516, Barcsoyssr_13_0687, Satt314, Satt338 and Satt448 and the absence of such at loci Satt229 and Satt006 are shown. According to the results of validation of markers of Cgy1, Cgy2, Cgdef genes encoding conglycinin subunits, the specificity of selected primer pairs to the genus Glycine was established by bioinformatics methods. Fragments of in silico PCR of the expected sizes were obtained for all genes. Another 250 bp amplification fragment was obtained for the Cgdef gene. According to DNA analysis at the Cgy-1 locus, 8 soybean varieties with the ability to accumulate conglycinin were identified. Analysis of the polymorphism of the Gy4 gene, which encodes the G4 (A4A5B3) subunit of glycine, revealed the presence of the allele of the "wild" type in varieties and samples of soybeans SGI-NCNS. 14 soybean varieties were identified by genes Gy1, Gy2, Gy3, Gy4, Gy5, encoding glycine subunits. The presence of a recessive or dominant allele of the E7 gene of photoperiodic sensitivity was determined for 15 soybean cultivars, based on the results of typing by the Satt100 microsatellite locus. For most samples, the correspondence between the presence of a certain allele of the E7 gene and the length of the growing season is shown.
