It was found that the adaptability of isolated hepatocytes depends on the method of isolation and is significantly higher upon in situ liver perfusion. Increasing the concentration of Ca2+ in the medium causes a decrease of the mitochondrial adaptive capacity in hepatocytes, and addition of CsA prevents the negative effect of high concentrations of Ca2+, but only if it was added before increasing the concentration of Ca2+. Ethanol in vitro did not affect maximal FCCP-stimulated respiration, mitochondrial membrane potential, and NADH fluorescence upon oxidation of glucose, pyruvate, and m-succinate. Chronic administration of alcohol to rats increases the mitochondrial adaptive capacity in hepatocytes, and in vivo administration of pyruvate does not affect it. In contrast, the administration of alcohol to rats adversely affects mitochondrial function, secretory response, and viability of pancreatic acinar cells, and pyruvate administration negates these effects. Administration of pyruvate to rats substantially ameliorates changes in the amino acid composition of plasma in models of acute pancreatitis.
