Information × Registration Number 0221U103943, 0118U005478 , R & D reports Title To determine the prognostic significance of intercellular interaction mediators and differentiation clusters of substrate cells for stratification and optimization of the myelodysplastic/myeloproliferative neoplasms treatment popup.stage_title Head Tretiak Naliia M, Registration Date 28-02-2021 Organization State Institution "Institute of Hematology and Transfusiology NAMS Ukraine" popup.description2  The object:61 patients with myelodysplastic/myeloproliferative neoplasms(MDS/MPN)aged 40-79 years.Objective:to optimize the criteria for predicting the effectiveness of therapy or disease progression based on the study of clinical and hematological,cytogenetic parameters,assessment of changes in markers of differentiation of bone marrow(BM)substrate cells(SC)and the effectiveness of colonies and clusters of BM progenitor cells in vitro in MDS/MPN which are treated with cytostatic drugs.Research methods:general clinical,immunophenotyping (flow fluorocytometry),cytological,cytogenetic,enzyme-linked immunosorbent assay,statistical.Subject:BM,BC,serum in 32 patients with MDS RAEBI and 29 patients with MDS RAEBII.The analysis of clinical and laboratory data and results of immunological examination of 61 patients with MDS/MPN in the initial period and after treatment with cytostatic drugs(CD).The immunophenotypic profile of BM tumor cells was determined by flow cytometry,the efficiency of formation of colonies and clusters of progenitor cells in Vitro culture,changes in the cytokine profile and the dynamics of expression of markers of differentiation of BM SC were studied.The tendency to prevail is revealed the relative number of myelocytes in the PC in MDS RAEBI,compared with RAEBII,most likely due to the greater number of maturing cells of the myeloid sprout.Hematological compensation in patients with MDS RAEBI obtained as a result of treatment with purinetol was maintained for 12-16 weeks,during which the need for hemotransfusions did not arise.As a result of cytosar treatment of patients with MDS RAEII 13% achieved hematological compensation,30% of patients stated stabilization of the pathological process(PP),13.4% of patients progressed the PP to transformation into AML.As a result of culturing BM cells of patients with MDS treated with CD,there was a pronounced inhibition of the colony-forming ability of progenitor cells compared with BM of healthy donors.
