The high-performance SPR spectrometer is designed to simultaneously perform many biological analyzes, it is a convenient tool for probing a large number of biomacromolecules and studying their interactions with various partner molecules. Surface plasmon resonance spectrometry was used to study the effectiveness of the process of immobilization of thiolated oligonucleotide fragments of the native or mutated rpoB gene of M. tuberculosis on the sensor surface of the DNA sensor and their hybridization with fully complementary targets. To assess the level of selectivity of bioselective elements formed using thiolated oligonucleotides HS-РNt or HS-Р2t, measurements of hybridization levels of immobilized oligonucleotides with both fully and partially complementary targets TNt, T2t and T3t were performed. The largest difference in responses was observed during hybridization of T2t and T3t targets with the HS-P2t probe, namely between interactions of fully complementary oligonucleotides and such partially complementary oligonucleotides containing two non-complementary nucleotides. For further use in the framework of this project, the configuration of the flow cell of the SPR spectrometer with a sensor chip in the format of a microarray with rounded corners was proposed.
