The work involved cytological, immunological, biochemical and statistical methods of analysis. The aim of the work was to develop a holistic system for assessing the course of the tumor process on the model of multicellular tumor spheroids. Based on the research results, a list of methods and approaches was identified that allow, in our opinion, to fairly objectively assess the state of the tumor population, the presence of a subpopulation of tumor stem cells, and the promising cytotoxic effect of potential anticancer drugs. A list of cytological, molecular, biochemical and immunological methods and guidelines were drawn up. Also, a stage of biochemical study of the state of the redox system of tumor cells under various cultivation conditions was implemented. The processes of peroxidation and neutralization of free radicals in BPS under the condition of their enrichment with OCP are suppressed compared to unenriched BPS. This may indicate a less “aggressive” environment in terms of oxidative stress of the SEC. Intensification of pentose phosphate shunt reactions in BPS enriched with OCPs can be an adaptive response to protect against excessive oxidative stress, and inhibition of glycolysis, which is probably a consequence of partial normalization of oxidative phosphorylation reactions in mitochondria, as a result of which glycolysis ceases to play a vital role in cells growing in this culture. Scientific and practical significance. As a result of this work, a wide range of methods for assessing the state of a tumor micro-node was formulated and tested, which allows one to proceed to the stage of obtaining a primary culture of tumor cells from the postoperative material of patients for generating BPS and assessing the effectiveness of anticancer drugs, targeting tumor stem cells, and setting up a clinically suitable test system.
