Yanovsky National academy of medical sciences of Ukraine” popup.description2 The object of study − strains of non-tuberculous mycobacteria (NTMB) from patients with mycobacteriosis of the lungs. The aim is to increase the efficiency and standardize methods of laboratory diagnosis of mycobacteriosis by developing an algorithm for gene-phenotypic diagnosis of pulmonary lesions caused by NTMB. Methods − genetic, bacteriological, statistical, equipment − phenotypic system BACTEC MGIT 960 and molecular genetic systems. It is established that in Ukraine 2.9 % of clinical isolates of mycobacteria are identified as NTMB. M. avium-complex was most often isolated − 45.5 %. A database of NTMB strains circulating in Ukraine has been created. The technology of gene-phenotypic diagnostics of pulmonary lesions caused by NTMB has been developed, which includes the optimal diagnostic complex for determining the species spectrum of NTMB strains. It is proved that for effective isolation of NTMB it is necessary to carry out sowing of experimental material on two nutrient media: liquid and dense. To accelerate the species identification of mycobacteria, the developed method of genetic identification of NTMB should be used, which allows to reduce the term of NTMB identification from 4 weeks to 2 working days. Standard operating procedures for laboratory diagnosis of mycobacteriosis have been created. External quality control of bacteriological tests for mycobacteriosis in Ukraine has been introduced. It is proved that the method of double serial microdilutions of drugs to determine the drug resistance of NTMB is the most promising for laboratories of anti-tuberculosis institutions in Ukraine.
