Purpose: to define the effectiveness and therapeutic activity mechanisms of bone marrow mesenchymal stromal cells in skin radiation lesions in rats on allogeneic and xenogeneic models. Methods and equipment: radiobiological, hematological, cell culture, morphological, statistical; CLINAC 6 MeV linear accelerator, UNIDOS universal dosimeter, MN 120 lamina shafts, EC-160 CO2 incubator, microscope Leica 6000B LED, chambers Leіca DFC 310 FX, microscope Leіca DM ІL LED, microscope Axіolab 94D 2601, analytical scales. Theoretical and practical results: the experimental model of severity skin radiation damages with chronic ulcers after local irradiation of rats thigh of rats at a dose of 75 Gy for testing allogeneic and xenogeneic therapeutic administration of cultured/cryopreserved BM MSC was developed. The viability of cultured/cryopreserved BM MSC intended for therapeutic administration was about 80% and their adipogenic and osteogenic differentiation ability were determined. It was found that the twice local skin injection of BM MSC at days14 and 21 after irradiation reduces the severity of local inflammation and clinical reactions in the skin irradiated area with accelerations of their healing by reducing the affected area by 2.6 times without radiation ulcers. It is determined that the systemic administration of cultured/cryopreserved MSCs KM (single or double) has a less therapeutic effect. Novelty: for the first time on the developed model of the skin radiation damages in rats the therapeutic efficacy of cultured/cryopreserved bone marrow mesenchymal stromal cells was evaluated.
