The possible promoter region of the MRPS18-2 gene was identified - at 600 bp. upstream from the ATG start codon. Analysis of the TRANSFAC database in the promoter region of the MRPS18-2 gene revealed three sites of possible binding of the transcription factor KLF4 protein and one for KLF6. KLF4 protein is involved in the regulation of somatic cell proliferation, differentiation, apoptosis and reprogramming. KLF4 protein is a suppressor of tumor growth, but in some nosological forms, such as Ewing's sarcoma, overexpression of the KLF4 gene is observed. KLF4 protein expression is high in embryonic and mesenchymal stem cells. The interaction of KLF4 transcription factor with the promoter region of the MRPS18-2 gene was confirmed by the chromatin immunoprecipitation. The dependence of MRPS18-2 gene transcription activation on the presence of KLF4 was shown, using quantitative PCR. Thus, the bioinformatics analysis of a possible promoter region of the MRPS18-2 gene revealed three sites for binding of KLF4 transcription factor and one for KLF6, which are important for maintaining of cell stemness and play an important role in malignant transformation and tumor progression. The dependence of transcription activation of the MRPS18-2 gene on the presence of KLF4, which induces cell pluripotency, partly explains the high levels of MRPS18-2 in stem and tumor cells.
