The genetic construct encoding a blood clotting activator has been developed. The prokaryotic and eukaryotic expression systems have been developed to obtain a protein coagulation activator. Laboratory regulations for obtaining recombinant protein coagulation activator and a protocol for isolating recombinant protein activator from culture medium, its refolding, concentration and storage have been developed. The procoagulant activity of the obtained recombinant protein coagulation activator was successfully tested. The procoagulant activity of recombinant protein coagulation activator under conditions of its combination with biomaterials to give them hemostatic properties has been studied and its high efficiency in vitro and in vivo has been shown. Based on the research, a protocol for the use of recombinant protein coagulation activator to stop bleeding alone or in combination with biomaterials was developed. Preclinical studies of protein coagulation activator have been successfully performed, namely the assessment of local tolerability of the drug based on natural and recombinant coagulation activator in accordance with OECD GLP requirements.
