Section 3. Development of reference methods for the arginine analysis with the usage of arginine-hydrolyzing recombinant enzymes. popup.stage_title Head Sybirnyi Andrii A., Registration Date 23-01-2022 Organization Institute of Cell Biology popup.description2  A number of electroactive nanomaterials with pseudo-peroxidase activity have been synthesized. The best artificial peroxidases were characterized amperometrically on the graphite electrode as sensors on H2O2 and used in the composition of amperometric biosensors (ABSs) on arginine (Arg). A partially purified preparation of arginine oxidase (ArgO) of the forest fungus Amanita phalloides was used as an Arg-sensitive element. As a result of research in 2020 year, new chemosensors for ammonium ions were proposed and ABSs for Arg was constructed, which contained recombinant bacterial arginine deiminase (ADI) and Cu / Zn (Hg) S nanoparticles. At the stage of 2021 year, the conditions for constructing this ADI-ABS were optimized, which led to a significant increase in its sensitivity to Arg. The created ADI-ABS and ArgO-ABS were tested on model solutions and real samples of juices in comparison with the reference enzymatic-chemical methods developed by us earlier. The close correlation of the results of Arg concentration’s determination in juice samples by biosensor and reference methods was demonstrated. Product Description popup.authors Gayda Galyna Z.
