The aim of the study was to investigate the effect of mutations in the BRCA1 gene on the development of genomic instability in human peripheral blood lymphocytes. Research methods: molecular genetic - polymerase chain reaction method; comet assay method; cytogenetic. A comparison of the level of genomic instability in irradiated peripheral blood lymphocytes of 10 people from the control group and 5 people - asymptomatic (without implemented oncopathology) carriers of the mutation in the BRCA1 gene. The peculiarities of the development of genomic instability in the carriers of the 5382insC mutation in the BRCA1 gene were revealed: a statistically significant (p <0.001) increase in the frequency of aberrant cells in irradiated peripheral blood lymphocytes of the carriers of the 5382insC mutation in the BRCA1 gene compared with 33 control groups ± 2.53 and 19.44 ± 1.22 per 100 cells, respectively), which indicates the chaotic repair processes, and, as a consequence, the increase in chromosomal instability in heterozygous carriers of the mutation; after irradiation, a decrease in the frequency of cells with a high level of DNA damage in the lymphocytes of peripheral blood of individuals carrying the 5382insC mutation in the BRCA1 gene (12.98 ± 2.33 vs. 38.74 ± 3.52, p <0.05), statistically significant increase in the frequency of cells in the state of apoptosis (from (5.95 ± 1.24) to (14.39 ± 2.16) per 100 cells per group on average, p <0.05). It was found that in violation of repair processes due to the presence of a hereditary mutation of 5382insC in the BRCA1 gene, apoptosis is a key mechanism for regulating the development of genomic instability in both unirradiated and irradiated somatic cells of asymptomatic carriers of mutations in the BRCA1 gene Product Description popup.authors Verbylenko Roksana M Demchenko Olena M.
