Research aim was to investigate the peculiarities of ultralow temperatures effect on molecular mechanisms of implementing the tolerogenic activity of dendritic cells (DCs) for optimization of cryopreservation and further establishing the banks for their preservation and adoptive usage in clinical practice when treating autoimmune diseases. Research methods were as follows: assessment of structural and functional characteristics of mononuclears and dendritic cells after cryopreservation by means of culturing in vitro, inverted and light microscopy, flow cytometry and real time PCR. Owing to theoretical grounding the method for cryopreservation of bone marrow MNCs under 10% DMSO protection with the rate of 1 deg/ min down to -40ºC with following plunging into liquid nitrogen, which provided structural and functional integrity of the obtained from them ex vivo immature DC with activation of heat shock proteins hsp70 in them and expression of GILS gene, tolerogenic activity regulators. It has been established that the DCs obtained in culture with cryopreserved on the selected regimen MNCS had a maximum ability to elevated content and suppressor activity of T-regulatory cells with foxp3 gene activation and stimulation of the production of anti-inflammatory cytokine IL-10. This resulted in maximal decrease of cell manifestation of adjuvant arthritis in animals, that explained molecular mechanisms of tolerogenic activity of cryopreserved dendritic cells during their adoptive application.
