Furthemore, development and optimization of the CRISPR/Cas9 protocol for genome editing and creation of high-precision deletions in genes of interest will be conducted. The project priority of this project is possibility to estimate the effectiveness of the CRISPR/ as9 approach in the generation of deletion knockoutsas well as evaluation of potential for the further application in plant biology, biotechnology and plant bioengineering. The maine task of the project second stage was to design primers for RNA-guides. Cloning and development of functional CRISPR / Cas9 constructs for generating deletions in genes encoding potassium channels NtTPK1a and NtTPK1b. Transformation of agrobacteria. As a result of the project implementation, the genome editing technique will be developed and optimized using the CRISPR / Cas9 system. Knockout lines of tobacco plants with deletions in the sequences of genes encoding TRC channels will be created. The created mutant plant lines will help to elucidate the physiological functions of the NtTPK1a and NtTPK1b channels in tobacco and assess their role in the response to abiotic stress, namely osmotic shock, salinity, and potassium deficiency.   Product Description popup.authors Isayenkov Stanislav V.
