Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2  The aim of the 2022 stage was to improve the manufacturing protocol of cytokine-induced killer cells (CIKs) and evaluate the effect of B. subtilis IMB B-7724 lectin on the chemo sensetive and chemo resistance breast cancer cell lines. As a result of the improved the manufacturing protocol of CIKs, a cell product with 80% content of functionally active CD3+CD56+ CIKs against tumor cells (NK-sensitive cell model of myelogenous leukemia) was obtained. The dose-dependent effect of B. subtilis IMB B-7724 lectin on tumor and normal human cells was shown. It has been proven that lectin at 0.1 mg/ml dose causes the death of 48-55% of MCF-7 tumor cells, regardless of their chemo sensitivity, and 38% of lymphocytes; at 0.05 mg/ml dose it increases the proliferative activity of peripheral blood lymphocytes, and at a dose of 0.02 mg/ml, it exhibits cytostatic activity against chemo resistant, but not chemo sensitive MCF-7 breast cancer cells. The obtained data confirm the expediency of further research on the combined use of CIKs and bacterial lectin on breast cancer cell lines with different malignancy grate and chemo sensitivity.
