The main objectives of the project: - to develop methods for the synthesis of new viscosity-sensitive fluorescent dyes based on squaraines; - to investigate the spectral and photophysical properties of the obtained dyes in solvents with different viscosities and polarities; - to find out the influence of structure and substituents on the spectral-luminescent properties of squaraine dyes and their sensitivity to viscosity. Objects of research: squaraine and cyanine dyes. Research methods: organic synthesis, spectral and other physical and photophysical research methods - NMR, electron absorption and fluorescence spectroscopy, mass spectrometry. The project results in a series of new fluorescent squaraine dyes with a terminal indolenine (3,3-dimethyl-3H-indolium) moiety and a secondary or tertiary amino group. The spectral and photophysical properties of the obtained compounds in solvents with different viscosities and polarities were studied. It was found that the transition from methanol to glycerol increases the quantum yields of dyes by 55-210 times. Non-covalent binding of synthesized squaraines to bovine serum albumin (BSA) also results in an increase in quantum yields due to changes in the polarity and viscosity of the microenvironment. The dependence of the spectral characteristics of the obtained dyes on the viscosity and polarity of the microenvironment makes them promising for testing as sensitive to viscosity probes in biomedical research.
