Karazin Kharkiv National University popup.description2  Methods of creation were developed and nanosized liposomes of a certain lipid composition with different concentrations of GS and different order of addition of components were obtained. The influence of different concentrations of GS on the viability, adhesive, proliferative and migratory properties of cells of the L929 line during 2D cultivation (in a monolayer) was studied. The concentration dependence of the effect of the antibiotic GS on the proliferative activity of cells was established: high synthetic activity and stimulating effect of the antibiotic GS on cell proliferation of fibroblasts in a monolayer were shown at concentrations of GS (0.5-5) μg/ml. At a concentration of 50 μg/ml, GS exhibits lytic activity against the L929 cell line culture. Addition of 5 μg/ml GS to the nutrient medium during monolayer formation induces the development of two cell populations in the culture - typical and cells that differ morphologically due to a large nucleus and increased cell area. To determine the influence of different concentrations of GS on the adhesive and migratory properties of L929 cells, the effect of GS on the properties of fibroblast L929 cells was investigated in vitro using the "scratch" test. Cells in culture are able to migrate to an area with lower cell density. At the same time, the recovery of the cell monolayer defect occurs to a greater extent due to cell migration and to a lesser extent to proliferation. It was established that GS in concentrations of 0.5 and 5 μg/ml does not affect the migratory and proliferative properties of cells of the L929 line, and in a concentration of 50 μg/ml inhibits them during short-term incubation for 1 hour. According to the results of research, the mechanism of interaction of the peptide antibiotic GS with biological membranes is determined and ways of optimizing the use of GS are determined, new potential targets of its action and the prospects for creating new dosage forms or methods of its encapsulation and d Product Description popup.authors Kuzeva Olga V.
