The protective mechanisms of H2S action can be related to both the restoration of constitutive Ca2+-dependent NO synthesis, the suppression of oxidative stress, and the effect on a non-specific megachannel - the mitochondrial transient conductance pore (mPTP). The use of PLP inhibited Ca2+-induced mPTP opening due to a decrease in its sensitivity to the inducer, which is significantly increased in old rats, and improved the processes of mitochondrial respiration. It was also shown that an increase in the content (by 53.6%) of the reduced form of glutathione in the heart tissue of old rats occurs after the administration of this antioxidant in a dose of 52 mg/kg. Under the conditions of action of exogenous glutathione in old animals, the activity of mtNOS, as well as the conjugated state of this enzyme, was restored. The introduction of exogenous glutathione significantly increased the content of total glutathione due to its reduced form, which led to the normalization of the redox status of mitochondria (reduction of oxidative stress), restoration of the activity and conjugation of mtNOS and H2S content, which can be manifested in cardiopotection. It was found that stimulation of endogenous synthesis of glutathione during aging increased the mitochondrial membrane potential and improved the efficiency of the respiratory chain. In the heart of old rats, the redox balance and nitric oxide metabolism were normalized. That is, with aging, endogenous stimulation of the synthesis of reduced glutathione by PAG +L-cysteine has a powerful cardioprotective effect.
