Research object: stem tumor cells, tumor spheroids of the MCF-7 line, monolayer culture of the MCF-7 line, doxorubicin, methotrexate, cisplatin. Cytological, biochemical, molecular biological methods and statistical methods of analysis were used in the work. The aim of the study was to compare the energy level, receptor and resistance state of the breast cancer cell population when the latter is enriched with stem tumor cells under the influence of microenvironmental factors and when 3D structures are organized. Determination of morphological and phenotypic characteristics of spheroids and monolayer culture under the conditions of incubation with antitumor drugs. The objectives of the study were to investigate the profiles of a subpopulation of tumor cells in mammospheres. To determine the influence of the conditions of the cellular microenvironment on the population of SPK in mammospheres, spheroid and monolayer culture (cytofluorometry). As a result, it was found that during the transition of adhesive cells to substance-independent growth, ER expression decreases by 52%, EGFR is expressed in 95% of all tumor cells in monolayer culture, while in spheroid culture only 25% of cells are positive for this marker. Cytokeratin receptors of clones AE1/AE3 and Pan in monolayer culture are represented in almost all cells, while in BPS the proportion of SC-positive cells was 15% and 45%, respectively. The percentage of EpCAM-positive cells was reduced by 72% in BPS compared to 2D culture. A 10% increase in the expression level of vimentin in spheroid cell culture compared to monolayer cell culture correlates with a decrease in the expression of estrogen receptors, cytokeratins, epithelial adhesion molecule receptor, and epidermal growth factor. Doxorubicin has the greatest cytotoxic effect on MCF-7 cells both in monolayer and spheroid cultures. On the other hand, methotrexate had Product Description popup.authors Baresh Oleksandr M.
