The purpose of the work is to develop methodical approaches to the creation of a bank of somatic cells of animals. Research methods - veterinary (tissue biopsy), general laboratory techniques, in vitro cell culture methods, cryobiology methods. Methodical approaches for taking tissue biopsies from animals, transporting and storing biopsies, disaggregating them and setting up a primary cell culture have been developed. The method of culturing and passage of cells has been developed and the acceptable number of passages for cell multiplication to the amount sufficient to create the necessary supply of them in the cryobank has been determined. Methodical approaches to cryopreservation of cells in cryostraws with a volume of 0.25 ml, their deconservation and systematic storage in a cryobank have been developed. Methodical approaches to in vitro culture and passage of cells after deconservation have also been worked out. Experiments to determine methodical approaches to the creation of a bank of somatic cells of animals were conducted on tissue biopsies taken from the ear skin of 3 ewes, 1 ram, 2 bucks, and 2 goats.
