The aim of the work was to study the effect of cryopreservation on placental cells (PCs) and to determine their impact on the reproductive system of females with ovarian failure (OF). Cultural, cytological, spectrophotometric, biochemical, histological, methods of light and electron microscopy, morphometric, statistical methods were used to carry out the work. As a result of the work, it was shown that the largest number of viable PCs can be isolated by the enzymatic method using trypsin, DNAase, ETDA, as well as the explant method. Incubation with pharmacopoeial infusion solutions, plasma substitutes and classic cryoprotectants in concentrations, determined for the preservation of other types of eukaryotic cells, does not lead to changes in the number and viability of PCs. The inclusion of slow freezing to –80°C in the PCs cryopreservation protocol at the first stage, followed by fast freezing to –196°С, significantly improves indicators of their preservation, vitality, metabolic activity and adhesive properties. The revealed nature of cryopreserved PCs (CPCs) effect in an in vitro system, namely, changes in the structure of uterus/ovaries organotypic cultures under the influence of CPCs conditioned medium, are similar to the changes occurring in vivo during pregnancy, which indicates the preservation of a heterogeneous pool of CPCs and their secretory activity. The use of CPCs in vivo has a positive effect on the general condition, physical parameters, structure of reproductive organs, contributes to the restoration of the estrous cycle and sexual function, increases life expectancy and survival of female mice in a model of chemotherapy-induced OF.
