In addition, a vector molecule model was developed based on a binary expression vector for cloning heterologous pHT43 proteins and previously obtained specific fragments of pag, lef and cya genes. Recombinant clones containing pHT43pag, pHT43lef and pHT43cya inserts were obtained on the basis of the shuttle vector pHT43 and Top10 E.coli strain. They carry gene fragments responsible for the synthesis of protective antigen (PA), lethal (LF) and oedema factors (EF) of the anthrax pathogen. Taking into account the safety, unpretentiousness and study of E. coli, microbial cells of recombinant strains containing specific fragments of immunogenicity epitopes can be offered as a basis for work on obtaining recombinant producer strains based on the more promising for these purposes B. subtilis as the main component of the recombinant vaccine against anthrax. It is assumed that B.subtilis is able to provide the production of immunologically active PA, LF and EF in amount that is not inferior to anthrax vaccine strains. On the basis of the work carried out during the reporting period, laboratory regulations and methodological recommendations for obtaining recombinant strains of E.coli expressing epitopes of the anthrax antigen were developed.
