The object of research is stem tumor cells, tumor spheroids of the MCF-7 line, monolayer culture of the MCF-7 line, doxorubicin, methotrexate, cisplatin, salinomycin. Cytological, immunocytological, molecular biological methods and statistical methods of analysis were used in the work. The purpose of the research is to compare the effects of salinomycin and classical anticancer drugs on breast cancer cells under the conditions of monolayer and spheroid growth according to several parameters. Determination of morphological and phenotypic characteristics of spheroids and monolayer culture under the conditions of incubation with antitumor drugs. The objectives of the study were to investigate the effect of antitumor drugs and salinomycin on the profile of a subpopulation of tumor cells in culture, to determine how salinomycin affects monolayer and spheroid cultures of MCF-7 cells. Compare the effect of salinomycin with the effect of classic anticancer drugs (doxorubicin, methotrexate, cisplatin). As a result, it was demonstrated that salinomycin causes a significant decrease in the survival and clonogenic potential of MCF-7 tumor cells in 3 monolayer culture compared to methotrexate, doxorubicin and cisplatin. It turned out that the survival of tumor cells in the conditions of monolayer and spheroid growth is significantly different. At the same time, for doxorubicin, methotrexate and cisplatin, there is an increased resistance of tumor cells in the conditions of spheroid growth to the cytotoxic effect of the drugs compared to monolayer culture. At the same time, when incubated with salinomycin, the spheroid culture was more sensitive to cytotoxic effects and showed a higher percentage of dead cells, compared to the monolayer growth model.
