Head Pirko Yaroslav V., Доктор біологічних наук Registration Date 02-12-2024 Organization Institute of Food Biotechnology and Genomics of the National Academy of Sciences of Ukraine popup.description1 The aim of this project is to develop breeding material of winter common wheat with the marked genes conferring resistance against highly pathogenic races of steam rust – new European races and the races of the Ug99 group to ensure food safety of Ukraine popup.description2  F3 plants from the crosses DH-G76-54 х Zymoyarka, DK20 x Samurai, DK20 x Solomiia, DK20 x Zymoyarka, Khutorianka x RL6089 and F4 plants from the cross DK20 x Myrkhad, which had been previously selected using molecular markers for the Sr33 or Sr40 gene, were grown. The lines with the Sr33 gene showed high resistance against the natural background of stem rust and leaf rust under conditions of 2024. Among winter lines derived from crosses with spring Canadian wheat material, genotypes with pseudoblack chaff (PBC), a morphological marker for the adult race-nonspecific stem rust resistance gene Sr2, were revealed. The presence of this gene was confirmed with the molecular marker csSr2. Using PCR analysis we performed marker-assisted selection of carriers of the Sr33 gene among the grown material and carriers of Sr39 in the additional sample of F4 spikes of the cross Solomiia x RL5711. The winter wheat material F4 and F5 selected with molecular markers was sown on the experimental plot. Seeds from F5 spikes from the cross DH31 x Myrkhad, in which the presence of the Sr33 gene had been presiously identified using the meolecular marker Sr33A, were also sown to develop F6 lines. Among the parental forms, the resistance allele of the Sr57 gene was detected in the lines DH-G76-54 and DH31. Using the markers caISBP1 and caSNP12, we identified carriers of the resistance allele of the Sr57 gene among the lines from the crosses DH-G76-54 х Zymoyarka and DH31 x Myrkhad, which had been previously selected for the presence of Sr33. The frequency of carries of the combination of two resistance alleles was low in the material from both crosses: 29 і 11%, respectively. Analysis of the previous material of ancestral F4 lines from the cross DH31 x Myrkhad, which included both genotypes with Sr33 and without it, revealed that the resistance allele of the target gene Sr33 was predominantly associated with the susceptibility allele of the Sr57 gene.
