The purpose of the work: to create an experimental model of human stem cells in vitro, to determine the viability of stem cells of various origins (survival, proliferative and mitotic activity), and to investigate the effectiveness of stem cell immobilization in the temporary equivalent of the dermal layer of the skin using hydrogel. Research methods: cultivation of human stem cells; light microscopy to assess the qualitative and quantitative characteristics of cells; determination of cell viability by staining with trypan blue; development and cryopreservation of biomaterials (cells of established lines and cell-free media conditioned by cells in culture (BCS); preparation and adaptation of cell carriers, hydrogels and membranes from materials of natural and artificial origin; creation of combined bioconstructions (carriers containing cells or BKS), methods of statistical data analysis.As a result of the work, new biotechnological products were created: a) using cell technology, equivalents of the dermal layer of the skin were obtained with the inclusion of human cells of the original lines or their derivatives (conditioned media containing a complex of biologically active substances synthesized by cells in culture); b) it is shown that dermal coatings based on a collagen membrane ("Bilkosin" film) and gelatin hydrogel containing immobilized stem cells or a derivative of these cells, BCS, can be the most effective bioconstructions that contribute to accelerated wound healing.
