Determination of anti-inflammatory and anti-amyloid activity of the drug, its effectiveness in relation to the dose and duration of action and aftereffects in vivo popup.description2 As part of the project, a new agent was created—an aerosol of lipid-targeted systems containing curcumin and microRNA for the treatment of neurodegenerative diseases. The results showed the anti-inflammatory and anti-amyloid activity of the agent, its effectiveness in terms of dose and duration of action, and after-effects in vivo. Samples were created in three basic concentrations of ingredients for further experimental in vivo research. The percentage of curcumin (98%) and miR-101 (96-98%) loaded into liposomes, their localization in liposomes, and the stability of liposomes during spraying were determined. An experimental model of the early presymptomatic stage of Alzheimer's disease in rats was created by intrahippocampal single administration of β-amyloid peptide_Human Ab42 aggregates. The dynamics of neuroinflammation development in the hippocampus and neocortex of the brain of animals with a model of Alzheimer's disease were shown. Intranasal testing of three doses of liposomal curcumin aerosol and miR-101 in rats with Alzheimer's disease showed that the most effective dose for suppressing amyloidosis, neuroinflammation, and oxidative stress was 0.55 mM Cur + 1.25 nM miR-101. The effective duration of the course of aerosol liposomal complex administration in animals with Alzheimer's disease model was determined to be 15 days. The duration of the anti-inflammatory and anti-amyloidogenic effect of a 15-day course of liposomal aerosol complex containing curcumin and miR-101 in animals with Alzheimer's disease lasts at least 10 days after discontinuation of treatment. The studies conducted made it possible to comprehensively assess and prove the high effectiveness of liposomal drug therapy for Alzheimer's disease based on biochemical indicators in target areas of the rat brain (neocortex, hippocampus).
