The determination of galactose content in clinical practice is important for the diagnosis of galactosemia (both classical and Duarte forms), while lactose analysis is necessary for controlling the composition of food products intended for people with lactose intolerance and children with galactosemia, as well as for controlling biotechnological processes related to the production of dairy products. popup.description2 Various microorganisms and cap fungi were screened for their ability to synthesize galactose oxidase (GalO). The culture of the fungus Trametes sp. 5138 was selected as the most effective source of the enzyme and the optimal conditions for cell growth and GalO synthesis were investigated. A simple scheme for the isolation and purification of GalO was developed by fractional salting out of cell-free extract proteins, ion-exchange and hydrophobic chromatography. A recombinant E. coli strain capable of oversynthesis of (His)6-tagged GalO was constructed, but the target enzyme is synthesized in an inactive form and accumulates mainly in the form of an “inclusion body”. A small part of the expressed enzyme is still soluble (the resulting enzyme preparation exhibits a specific activity of ~ 3.6 μmol min −1 ·mg −1 protein). The possibility of obtaining functionally active nanocomposites containing synthetic nanoperoxidases and galactose oxidase has been shown, and their kinetic properties have been studied - both in solution and in an immobilized state on the surface of electrodes.
