Abstract: The data has been verified for direct differentiation of MSCs in monospherical (2D) and spheroidal (3D) cultures and on cluster blocks. The survival rate of bone marrow MSCs when cultured in different systems was determined: in monosphere (2D) and spheroid (3D) cultures and on bone blocks. The viability of MSCs was determined (based on cytometric data) after different cultivation schemes at the intervals of 1, 2, 3, 4 days of cultivation. The best result of proliferative activity was achieved for MSCs when cultured on cyst granules: an increase in the number of living cells by 105%. When incubated in a monospherical (2D) culture – by 78%, and in a spheroidal (3D) culture, changes within the framework of statistical loss. It has been established that MSCs grown on cystic granules retain pluripotency and ability to differentiate in different directions. At the same time, cells in rich cell spheroids demonstrate less formation before differentiation. When the differentiation medium is selected, which stimulates the adipo-differentiation of MSCs, the aggregates closest to the size, distribution and external morphology are selected to the control signs. Thus, the majority of MSC aggregates (45%) have a volume between 1.0 and 5.0 µm3. At the same time, in the chondro-differential middle, small spheroids with a volume of 0.1 to 0.5 µm3 prevail (26%). Cultivation of MSCs in osteodifferentiated media allows the recovery of MSC spheroids ranging in size from 1.0 to 5.0 μm3 in 40% of the normal population. CD73 expression was lowest in cells from 2D culture (54.7%) and highest in cells proliferated on cyst granules (85.5%). CD90 expression was highest in monosphere (66.3%) and spheroid cultures (67.65) and even lower in cyst blocks (51.6). CD105 expression was highest in cells that proliferated on disk granules (81.6%) and least expressed in spheroid cultures (78.6%) and least expressed in monosphere cultures (52.6%).
