The aim of work is to isolate DNA (deoxyribonucleic acid) from intact maize plants belonging to various lines differing from each other by their ability to regenerate in vitro and generate callus cultures from them followed by the regenerant induction. The method of investigation is plant tissue culture and cellular selection in vitro as well DNA isolation from plants. Plants of two maize lines differing from each other by their regeneration ability in culture in vitro were grown in the field. Their pure-line pedigree seed was obtained through self-pollination and the explants of the immature embryos for culturing in conditions in vitro were obtained. The optimal composition medium for induction of maize callusogenesis was devised. It is composed of base Murashige and Skoog medium [1] supplemented with 2 g/l 2,4-D and 30 g/l sucrose. Passaging callus cultures from immature embryos of 1-1.5 mm length and 12-14 days old after self-pollination were obtained. Plants-regenerants from primary calli of two maize lines differ from each other by ability to regenerate were obtained as well. Differences between lines by the "regeneration ability" trait was ascertained through the comparitive counts of the regenerants generated. The effective method of DNA isolation from maize plant material was found. It was established that DNA isolated from maize under method [2] was digested by restrictases and available for the future molecular-biology works. DNA samples from intact plants of the two investigated maize lines were isolated. Protocol for maize callusogenesis and induction of regeneration was done under results of the work (see Appendix A).
