Moreover, a new diagnostic kit "LACTATEST" for L-lactate analysis (using flavocytochrome b2 (L-lactate: cytochrome c-oxidoreductase) has been created at the Department of Analytical Biotechnology, Institute of Cell biology NAS of Ukraine. The developed methods were adapted to the analysis of the cultural liquid and cell-free extracts of tumors. The proposed methods were used to determine glucose and L-lactate in the cultural liquid (both substances) and cell-free extracts (L-lactate) of human myeloma, line RPMI 822. The detected production of L-lactic acid in the culture of myeloma confirms a Warburg phenomenon for this type of cancer cells. It was shown that bromopyruvate (inhibitor of glycolysis) in a concentration of 25 µM reduces the rate of glucose uptake. There is a tendency of decreased production of lactic acid in the culture of the human myeloma RPMI 822 as a consequence of the bromopyruvate treatment.
