The purpose of the study is to assay a cell culture system of multicellular tumor spheroids enriched with cancer stem cells (eMCTS) to determine the effect of anticancer drugs. Research methods of the study are cultivation of adhesive cells and multicellular tumor spheroids (MCTS), immunohistochemical, morphometric, spectrophotometric, cell survival analysis (MTT test), counting of the cells in Goryaev's chamber. Increasing expression of CSCs markers in the eMCTS culture was shown, compare with the adhesive culture of MCF-7 cells, which were cultured under standard conditions. It has been determined that the eMCTS culture is less sensitive to the effects of anticancer drugs (cisplatin, methotrexate and doxorubicin), compared with MCTS and monolayer culture. It has been established that cells can acquire stem phenotype and lose it depending on the conditions in which they are found. The processes of peroxidation and neutralization of free radicals in the eMCTS are suppressed in comparison with MCTS, that non-enriched with CSCs. Intensification of the pentose phosphate pathway and inhibition of the glycolysis reactions in cells of eMCTS, compared with cells of monolayer cell culture and MCTS, are also shown.
